چکیده

INTRODUCTION Alkaline agarose gels are run at a pH that is sufficiently high to denature double-stranded DNA. The denatured DNA is maintained in a single-stranded state and migrates through the alkaline gel as a function of its size. Alkaline agarose gels are used chiefly to measure the size of first and second strands of cDNA (Construction of cDNA Libraries Stage 1: Synthesis of First-strand cDNA Catalyzed by Reverse Transcriptase) and to analyze the size of the DNA strand after digestion of DNA-RNA hybrids with nucleases such as S1.

Cloning and expression of the bovine intestinal alkaline phosphatase gene: biochemical characterization of the recombinant enzyme.

A complete genomic clone and a full-length cDNA coding for bovine intestinal alkaline phosphatase have been isolated and sequenced. The gene (5.4 kb) contains 11 exons separated by ten small introns at positions identical to those other members of the eukaryotic tissue-specific alkaline phosphatase family. In addition, 1.5 kb of upstream sequences contain putative regulatory elements showing se...

Hemoglobin electrophoresis at alkaline pH on agarose gels.

We describe a modified thin agarose-gel system for use in primary hemoglobin screening by electrophoresis. The system makes use of a Tris-EDTA-borate-buffered agarose gel (pH 8.8) and a sodium barbital electrophoresis buffer (pH 8.6). Separation is effected in 20 min at a constant potential of 250 V. Eight samples can be simultaneously separated and the patterns made visible in 47 min. There ar...

Altered form of placental alkaline phosphatase produced by JAR choriocarcinoma cells in culture.

The alkaline phosphatase activity expressed by JAR choriocarcinoma cells was compared to the placental isoenzyme of human alkaline phosphatase by several criteria. JAR cell alkaline phosphatase was similar to the placental isoenzyme with respect to heat and urea stability and sensitivity to most inhibitors, but it differed significantly from placental alkaline phosphatase in its sensitivity to ...

A variant alkaline phosphatase found in a case of gastric carcinoma with super bone scan.

A rare case of gastric carcinoma associated with increased serum variant alkaline phosphatase activities is presented. A 54 year old man had extremely high serum alkaline phosphatase activity (18,607 U/l) with normal calcium and phosphate concentrations. His bone scintigram showed abnormal findings, 'super bone scan'. He was diagnosed as having Borrmann type 4 gastric carcinoma with diffuse bon...

Comparison of alkaline phosphatase from human normal and leukemic leukocytes.

The alkaline phosphatase of leukocytes from normal subjects, patients with chronic granulocytic leukemia, and patients with reactive granulocytosis have been characterized by means of agarose column chromatography, sucrose density ultracentrifugation, starch gel electrophoresis, Michaelis constant for p-nitrophenyl phosphate, heat inactivation, inhibition by L-phenylalanine, Ouchterlony double ...
